摘要	目的　被尖吻蝮蛇(D ienag k istrod on acu tus) 咬伤会引起严重的出血, 对蛇毒出血毒素的研究有利于治疗蛇伤出血药物筛选。方法　采用Sephadex G275, DEA E2Sephadex A 250, Sephadex G2200 和两次 PBE 聚焦层析纯化。SDS2PA GE 电泳和等电聚焦电泳测定纯化样品的纯度和等电点。氨基酸组成用自动氨基酸分析仪测定。以小鼠背部皮下注射部位出血斑的面积来确定最小出血剂量和常规的方法测定酶活性。结果　从尖吻蝮蛇毒中纯化到一个相对分子量为56 000 的出血毒素(DaHT23) , 经氨基酸组成测定计算, 它由487 个氨基酸残基组成。此成分在SDS2PA GE 上显示出一条均一的蛋白染色带, 其p I 为5150。该出血成分的最小出血剂量是216Lg, 具有蛋白水解酶活力, 其活力为3168, 但没有精氨酯酶和磷脂酶A 2 活力。当加入EDTA 螯合剂去除金属离子后, 它们的出血活力和蛋白水解酶活力均丧失。结论　这是从大陆尖吻蝮蛇毒中获得的一个新的出血金属蛋白酶(DaHT 23)。; O bjective 　Studies on hemo rrhagic tox in s of snake venom s are benef icial fo r medical screen ing1M ethod s 　F ive step separat ion s of Sephadex G275 f ilt rat ion, DEA E2 Sephadex A 250 ch romatography, Sephadex G2200 f ilt rat ion, and two Po lybuffer2exchang2 er ch romatofocu sing w ere emp loyed1 The pu rif ied samp le and mo lecu lar w eigh t w ere est i2 mated on SDS2po lyacrylam ide gel elect ropho resis1 p Iw as detected by the gel of isoelect ric focu sing elect ropho resis1 Am ino acid compo sit ion w as determ inated w ith am ino acid au to2 analyzer1 The m in imum hemo rrhagic do se (MHD ) w as def ined as the low est do se w h ich cau sed an area of hemo rrhage of 5mm in diameter 6 hou rs af ter subcu taneou s in ject ion1 The pu rif ied samp le w as assayed by u sing casein as sub st rate1 A rgin ine esterase act ivity and pho spho lipase A 2 act ivity w ere ob served w ith TAM E, BA EE and lecith in as sub2 st rates1 R esu lts　A new hemo rrhagic tox in (DaHT 23) w as pu rif ied f rom the venom of D i2 enag k istrod on acu tus1 DaHT 23 had mo lecu lar w eigh t of 56 000 w ith 487 am ino acid residues and isoelct ric po in t (p I) w as 51501 Them in imum hemo rrhagic do sew asmeasu red to be 216Lg1It exh ib ited p ro teo lyt ic act ive un it of 3168 and there w ere no argin ine esterase act ivity and pho spho lipase A 2 act ivity1W hen the hemo rrhagic tox in t reated w ith EDTA w as again assayed fo r hemo rrhagic and p ro teo lyt ic act ivity, the bo th act ivit ies disap2 peared1 The yield of DaHT 23 w as on ly 015% 1 Conclusion　Th is hemo rrhagic tox in w as a metallop ro teinase1
