on the salivary extraction of horsefly so far. So we chose horseflies——Tabancus yao Macquart as our material.We worked on tabancus yao Macquart to find some candidates for antithrombotic medicine. In order to get materials to study the functions and toxicity of the antithrombotic substances from Tabancus yao Macquart salivary in vivo, we expressed some of the antithrombotic substances at laboratory scale.
We screened the cDNA library of tabancus yao Macquart salivary. And the whole length sequence of tablysin2, a fibrinogenase of tabancus yao Macquart salivary, was found. Then we expressed it with the pET expression system. And tablysin2 with fibrinogen hydrolytic activity and platelet aggregation inhibitory activity was produced. This progress lifted the limitation of the deficiency in resources. And it made the study of tablysin2 in vivo and the clinical application of tablysin2 possible. Additionally, we studied the bleeding side effect of tablysin2 and found that tablysin2 didn’t cause bleeding under the concentration of 18ug/ml.
