Cathelicidins are a family of multi-functional antimicrobial peptides found in mammals, birds, reptiles and fish by far. Cathelicidin-TV is the first cathelicidin found in caudata of amphibia. Tylototriton verrucosus is a kind of tylototriton belonging to amphibia, caudata, salamandridae. It is widely distributed in south of Yunnan, Guangxi, north of Thailand and Southeast Asia region. In this thesis, a series of experiments were carried out to study cathelicidin-TV from T. verrucosus. A novel cathelicidin, named as cathelicidin-TV, was purified from skin of T. verrucosus by gel filtration and RP-HPLC. Edman degradation method was used for the determination of its amino acid sequence. The determined sequence was KCVRQNNKRVCK, composed of 12 amino acid residues. By electrospray ionization mass spectrometry (ESI-MS), the molecular mass of cathelicidin-TV was determined to be 1473.8 Da. A 438 bp cDNA sequence encoding the precursor of cathelicidin-TV was cloned from a cDNA library of T. verrucosus skin.
